How UV Plate quantifies DNA and Protein content in samples:

When 260nm short-wavelength ultraviolet light irradiates the DNA sample, it will be adsorbed by DNA molecules, and the transmittance of ultraviolet light will decrease. On the contrary, the optical density value (Optical Density) measured by the spectrophotometer will increase in contrast. The size of the density value can be calculated by the software program to calculate the actual content of DNA. When 280nm short-wavelength ultraviolet light irradiates the protein sample, it will be absorbed by the protein molecules

 In addition, the transmittance of ultraviolet light decreases, on the contrary, the optical density value (Optical Density) measured by the spectrophotometer will increase in contrast, and the actual content of DNA can be calculated by the software program by the detected optical density value.

The general 96-well clear plate is made of PSˎPCˎ or PET. When the light source passes through the 260nm filter and the screened beam is irradiated to the PSˎPCˎ or PET material interface, the short-wavelength ultraviolet light is absorbed by the medium and cannot pass through the interface, so it cannot be used as UV light plate to use.
